      目的 观察四味余甘子方对高脂血症模型大鼠血脂、CETP、FFA、ABCA1影响。方法 将SPF级SD雄性大鼠60只,体质量200g±20g,采用随机数字表法完全随机分为6组(n=10),正常对照组、模型对照组、血脂康组、四味余甘子方低剂量组、四味余甘子方中剂量组、四味余甘子方高剂量组,正常对照组予普通饲料,其余各组在予普通饲料基础上加脂肪乳剂灌胃。除模型对照组以生理盐水灌胃外,其余各组分别按人-动物药物剂量换算表予以相应剂量药物灌胃。4周后,采用ELISA法检测大鼠血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)的水平,同时采用ELISA法检测血清ATP结合盒转运蛋白A1(ABCA1)、胆固醇酯转运蛋白(CETP)、血清游离脂肪酸 (FFA)的水平,另采用免疫组化法检测大鼠肝脏组织ATP结合盒转运蛋白A1(ABCA1)的水平。结果 与正常对照组相比,模型对照组TC、TG、LDL-C明显升高,HDL-C、血清ABCA1明显降低(p<0.05),且肝脏ABCA1的表达亦降低(p<0.05),CETP、FFA升高(p<0.01);与模型对照组相比,血脂康组及四味余甘子方低、中、高剂量组TC、TG、LDL-C下降(p<0.05),HDL-C及血清ABCA1增加(p<0.05),且肝脏组织ABCA1的表达明显增加(P<0.05),CETP、FFA下降(p<0.01);四味余甘子方高剂量对血脂、ABCA1、CETP及FFA影响最大(p<0.05,p<0.01)。结论 四味余甘子方对高脂血症模型大鼠的血脂有调节作用,且能降低血清CETP及FFA,增加ABCA1的表达。

      OBJECTIVE: To observe the effect of Siwei Yuganzi Decoction on serum lipid, CETP and FFA in hyperlipidemia model rats and its effect on ABCA1 expression. METHODS: Sixty specific pathogen free male Sprague-Dawley rats, weighing 200g±20g, were randomly divided into 6 groups (n=10) using a random number table method,Normal control group, model control group, Xuezhikang group, Siwei Yuganzi Decoction low-dose group,Siwei Yuganzi Decoction middle-dose group,Siwei Yuganzi Decoction high-dose group. The normal control group was given normal feed, and the other groups were given intragastric administration of fat emulsion on the basis of common feed. Except that the model control group was intragastrically administered with normal saline, the other groups were given the corresponding doses of drugs by gavage according to the human-animal drug dose conversion table.Four weeks after administration, the levels of TC, TG, HDL-C, and LDL-C were measured by ELISA, and the levels of ABCA1,CETP and FFA were also measured by ELISA, and the levels of ABCA1 in rat liver tissue were measured by immunohistochemistry. RESULTS: Compared with the normal control group, the TC, TG and LDL-C in the model control group were significantly increased, HDL-C and serum ABCA1 were significantly decreased (p<0.05), and the expression of hepatic ABCA1 was also decreased (p<0.05), CETP and FFA were increased (p<0.01); compared with the model control group, the TC, TG and LDL-C in the Xuezhikang group and the low-, medium- and high-dose Siweiganzi group were decreased (p<0.05), HDL-C and serum ABC1 were increased (p<0.05), and the expression of hepatic ABCA1 was significantly increased (P<0.05), CETP and FFA were decreased (p<0.01); the high dose of Siweiganzi recipe had the greatest effect on blood lipid, ABCA1, CETP and FFA (p<0.05, p<0.01). CONCLUSIONS: Siwei Yuganzi Decoction can regulate blood lipid of hyperlipidemia model rats, and can reduce their seru CETP and FFA, increase their expression of ABCA1.
