目的 建立葡聚糖硫酸钠（Dextran sulphate sodium，DSS）诱导的小鼠肠炎模型，观察鸦胆子药渣提取物对溃疡性结肠炎（Ulcerative colitis，UC）小鼠的治疗作用。方法 采用DSS自由饮水建立UC小鼠模型，将BALB/c小鼠随机分为正常组、模型组、美沙拉嗪（5-ASA，200 mg·kg-1），鸦胆子药渣低剂量组（25 mg·kg-1），中剂量组（50 mg·kg-1），高剂量组（100 mg·kg-1），通过观察小鼠一般情况并评估疾病活动指数（DAI），结肠大体形态，组织学改变，以及通过ELISA法测定小鼠结肠组织TNF-α、IL-1β、IFN-γ水平。结果 鸦胆子药渣能够明显改善DSS导致的小鼠体重下降，DAI评分和组织损伤评分的升高；以及通过鸦胆子药渣各剂量治疗后，小鼠结肠组织中TNF-α、IL-1β、IFN-γ水平较模型组均显著下降。结论 鸦胆子药渣能有效缓解UC模型小鼠结肠炎症，其机制可能与抑制促炎症细胞因子TNF-α、IL-1β、IFN-γ水平升高有关。
Objective To evaluate the effects of extraction from dregs of Brucea fructus on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in mice. Methods BALB/c mice were randomly divided into 6 groups: a normal control, a DSS group, a positive groups (5-ASA, 200 mg·kg-1) and extraction-treated groups (25, 50, 100 mg·kg-1). The DSS group was fed on 3% (v/w) DSS for 7 days while clean water for normal group. The effects of extraction were evaluated by body weight loss, disease activity index (DAI), colon length, histological evaluations, and the levels of pro-inflammatory cytokines (TNF-α, IL-1β, IFN-γ). Results The body weight loss and colon length were significantly recovered by extraction. Meanwhile, the DAI and microscopic score of the group treated with extraction were lower than those of the model group. Furthermore, treated with extraction, the levels of TNF-α, IFN-γ, IL-1β were significantly reduced when compared with model group. Conclusion The extraction from dregs of Brucea Fructus could effectively attenuate murine experimental UC induced by DSS. The mechanism of extraction might be associated with its suppression of pro-inflammatory cytokines productions.
