目的 建立雅连不同炮制品的炮制工艺，测定炮制前后7个生物碱的含量，构建雅连科学的质量控制方法，以提升雅连药材、饮片、精制饮片的质量控制水平。方法 通过四因素三水平的正交实验，以生物碱总含量为指标，筛选了酒雅连、姜雅连、萸雅连的最佳炮制工艺；采用高效液相色谱法测定雅连不同炮制品总生物碱以及7个生物碱（小檗碱、黄连碱、巴马汀、药根碱、表小檗碱、非洲防己碱、格陵兰黄连碱）含量。经方法学验证，该方法精密度、重现性、稳定性均符合要求。结果 确定了雅连各炮制品的最佳炮制工艺；含量测定结果表明，萸黄连炮制后总生物碱含量最高，姜黄连次之，皆高于生品，7个生物碱含量，因炮制方法不同差别较大。结论 雅连各炮制品最佳炮制工艺的确定结合多指标成分定量，可以有效控制雅连药材的质量，为2020年版《中华人民共和国药典》黄连药材质量标准的完善与提高提供参考。
Objective To establish the processing technology of different processed products of Yalian, and to determine the content of 7 alkaloids before and after processing, and construct the scientific quality control method of Yalian to improve the quality control level of Yalian medicine, decoction pieces and refined pieces.Methods Four-factor and three-level orthogonal experiments were carried out to select the best processing technology of Jiu Lian, Jiang Yalian and Yu Yalian based on the total alkaloid content. The high-precision liquid chromatography method was used to determine the total biological products of different products of Yalian and 7 alkaloids (berberine, coptisine, palmatine, jatrorrhizine, epi-berberine, adenine, and greening berberine). The method has proved that the precision, reproducibility and stability of the method meet the requirements.Results The best processing technology of the products of Yalian was confirmed. The content of the samples showed that the total alkaloid content of the scutellaria chinensis was the highest, and the turmeric was the second highest, which was higher than the raw materials and 7 alkaloids.Conclusion The determination of the best processing technology of Yalian"s artillery products combined with multi-index component quantification can effectively control the quality of Yalian medicinal materials, and provide reference for the improvement and improvement of the 2020 edition of the Pharmacopoeia of the People"s Republic of China.
