目的 观察当归-川牛膝水煎剂对自发性高血压大鼠（SHR）血压、血清中内皮素（ET-1）、一氧化氮（NO）表达的变化。方法 6周龄48只雄性SHR随机分为8组，分别是模型对照组、缬沙坦组、当归-川牛膝组和分离组（水、30%乙醇、50%乙醇、70%乙醇和90%乙醇），每组6只，并将同周龄的WKY大鼠作为正常对照组，各给药组以灌胃方式给药，模型组和正常对照组给生理盐水，连续治疗4周。Elisa法检测血清中内皮素（ET-1）、一氧化氮（NO）表达水平。结果 模型组ET-1水平明显高于正常对照组，NO水平明显低于正常对照组，差异均具有统计学意义（P < 0.05）；缬沙坦组、当归-川牛膝组、30%醇洗组ET-1水平明显低于模型组，NO水平明显高于模型组，差异均具有统计学意义（P < 0.05）。结论 当归-川牛膝水煎剂及其提取物能有效降低SHR血清中ET-1水平，增加NO浓度，抑制血压升高。
Objective To observe the changes of the blood pressure, serum endothelin (ET-1) and nitric oxide (NO) expression in spontaneously hypertensive rats (SHR) treated with Radix Angelicae Sinensis-Radix Achyranthes Bidentata decoction.Methods Forty-eight male SHR patients aged 6 weeks were randomly divided into 8 groups: model control group, valsartan group, Radix Angelicae Sinensis - Radix Achyranthes Bidentata group andisolation group (water, 30% ethanol, 50% ethanol, 70% ethanol and 90% ethanol). WKY rats of the same age were taken as the normal control group, and each group was given by gavage. The model group and the normal control group were treated with normal saline for 4 weeks. The expression of endothelin (ET-1) and nitric oxide (NO) in serum were detected by ELISA method.Results The level of ET-1 in the model group was significantly higher than that in the normal control group, and the level of NO in the model group was significantly lower than that in the normal control group (P < 0.05). The levels of ET-1 and NO in valsartan group, Radix Angelicae Sinensis-Radix Achyranthes Bidentata group and 30% alcohol washing group were significantly lower than those in the model group, and the difference was statistically significant (P < 0.05).Conclusion Radix Angelicae Sinensis-Radix Achyranthes Bidentata and its extract can effectively reduce the level of ET-1 in serum, increase the concentration of NO and inhibit the rise of blood pressure.
